The Mag-Bind® Blood & Tissue DNA HDQ 96 Kit is designed for the rapid and reliable isolation of high-quality genomic DNA from 100-250 μL of blood samples, 500 μL saliva, swabs, mouse tails, dried blood spots, tissues, or 5 x 106 cultured cells. Mag-Bind® Particles HDQ provide quick magnetic response times, thereby reducing overall processing time.

Our cutting-edge Mag-Bind® technology reduces overall processing time by providing a quick magnetic response. This kit combines the reversible nucleic acid-binding properties of Mag-Bind® paramagnetic particles with the proven efficiency of Omega Bio-tek’s blood and tissue DNA isolation system to provide a rapid and robust method for the isolation of DNA from a variety of biological samples. The system yields high-quality DNA that is suitable for direct use in most downstream applications such as amplification, NGS, and enzymatic reactions.

Can the HDQ Binding Buffer and the Mag-Bind Particles HDQ be made into a master mix?

Yes, they can be made into a master mix before starting the experiment.

Do you have a protocol for dried blood spots using the Mag-Bind® Blood & Tissue DNA HDQ 96 Kit?

  1. Cut or punch-out the blood spot from the filter paper (up to 200 µL blood can be used per spot). Tear or cut the filter paper into small pieces and place them into a 2 mL nuclease-free microcentrifuge tube.
  2. Add 250 µL PBS.
  3. Incubate at 65 °C for 1 hour. Vortex briefly every 20 minutes.
  4. Add 25 µL Proteinase K Solution. Vortex at maximum speed for 15 seconds.
  5. Incubate at 65 °C for 30 minutes. Vortex briefly several times during incubation.
  6. Centrifuge at ≥13,000 x g for 5 minutes.
  7. Transfer the supernatant to a nuclease-free 2 mL microcentrifuge tube.
  8. Add 290 µL AL Buffer and vortex, or pipet up and down 20 times, to mix. Optional: Add 5 µL RNase A to each sample. Vortex to mix. Let sit at room temperature for 2 minutes.
  9. Continue from step 5 (page 6) of the ‘Blood Protocol’ (250 µL) on the Mag-Bind Blood & Tissue DNA HDQ 96 Kit manual.

Can the Mag-Bind® Blood & Tissue DNA HDQ 96 Kit be used for extraction of blood in PAXgene™ tubes?

Yes. See the protocol modification below. Please be aware we have not validated this protocol.

  1. Pour all the blood from one PAXgene blood DNA tube into a 50 mL centrifuge containing 25 ml NL buffer. Mix by inverting the tube 5 – 10 times.
  2. Centrifuge for 5 minutes at 2500 x g in a swing bucket rotor. Carefully discard the supernatant.
  3. Suspend the pellet in 1 mL of PBS or elution buffer.
  4. Follow the M6399 ‘Blood Protocol’ – either scale up the protocol 4 times or run 4 parallel preps of 250 µL each.

I accidentally left the magnetic beads out when it says to store them in the fridge. Are they ok to use?

The Mag-Bind particles are stable at room temperature for 2 weeks.
