Detecting Viable Fire Blight Pathogens in Apple Bark: How Improved DNA Extraction Enables Deeper Insight
Fire blight remains one of the most destructive diseases affecting apple production worldwide. Caused by the bacterium Erwinia amylovora, the disease persists in woody tissues known as cankers, which act as hidden reservoirs that fuel new infections each growing season. In the study “Improved Canker Processing and Viability Droplet Digital PCR Allow Detection of Erwinia amylovora Viable Nonculturable Cells in Apple Bark” (Microorganisms, 2024), researchers examined apple bark canker samples from ‘Honeycrisp’ and ‘Evercrisp’ trees and demonstrated how optimized sample processing and DNA extraction are essential for accurately detecting viable pathogens in complex plant tissues.
Traditional culture-based methods often fail to reveal the full picture of pathogen survival in cankers.
